Experimental Study on the Interrelation between Liver Regeneration and EGFR after Parital Hepatectomy in Rabbits


Abstract
The epidermal growth factor(EGF) is a polypeptide growth factor og 53 aminoacid that stimulate the proliferation of hepatocyte in vitro and in vivo. The action. of EGF is mediated activation of specific cell surface receptor(epidermal growth

The aim of this study was to elucidate interrelation between liver regeneration and EGFR after 75% partial hepatectomy(PH).
Fourty five healthy adult New Zealand white rabbits weighing about 2kg were divided into 2 groups: sham operation group as control(n=5) and 75% hepatectomy group(n=40, PH group)
In PH group, 5 rabbits were sacrificed at every 12 hours, 1st day, 2nd day, 3rd day, 7th day, 14th day, 21st day, and 28th day after PH.
The H-E staining, Ag-NOR stains, and immunohistochemical stains for EGFR were checked in PH group for elucidation of interrelation between liver regeneration and EGFR after PH.
@ES The results obtained were summerized as follows:
@EN 1. The mean regeneration ratio of residual liver was 27.7% at 12th hour, 36.8% at 1st day, 46.3%, at 2nd day, 54.7% at 3rd day, 83.8% at 7th day, 105.4% at 14th day, 116.95 at 21st day, and 116.2% at 28th day in PH group
2. The mitosis, dark cells, and double nucleated cells reached peak level at 2nd~3rd day and returned to control level at 21st day after PH.
3. The mean vlaue Ag-NOR index(number of NORs/100 hepatocytes)was 170.2¡¾5.7 ion control group. In PH group, it was 205.4 at 12th hour, 218.2 at 2nd day raching its peak, decreased slowly, and finnally returned to control level at 14th day after
PH.
4. The degree of numbers of EGFR positive hepatocytes in EGFR stains ws severe degree in control group. It was mild at 12th hour, negative or mild at 1st day, milder moderate at 2nd day, moderate at 3rd day, moderate or severe at 7th day, and
severe
degree at 14th day reaching control level in PH.
Based on these results, it is suggested that EGFR reduction of regenerating liver cells after partial hepatectomy may be closely related with mitosis and DNA synthesis of hepatocytes in association with liver regeneration.
